Metabolon – Innovative Diagnostics and Research Services through Advanced Metabolomics

The DHMRI Metabolomics Laboratory has partnered with Metabolon, an industry leader in metabolomics, to provide a range of metabolomic profiling services that quickly deliver actionable data for nutrition and obesity researchers.

Metabolon’s expert biochemists, molecular biologists and nutritionists work directly with collaborators to understand investigator research goals, design well-powered metabolomics studies, and interpret the data output from small and large sample sets to yield meaningful results. A publication-ready report and other deliverables convey study outcomes, provide statistical and biological interpretation and identify candidate biomarkers.


Working with Metabolon

Metabolon offers metabolomic profiling services on hundreds of different sample types from most species. Results are typically returned within six to eight weeks.  The number of metabolites identified depends on the sample type, but typically ranges from 500-1000 metabolites in common sample types such as urine or plasma.

Our metabolomics study process is simple, scalable, and client-centric to help investigators achieve their research goals:

Step 1: Consultation & Study Design -  Each study is assigned to an experienced Ph.D.-level project manager, who works with clients to design a study that precisely achieves their objectives. Study design consultation is included in most all service offerings, and there is no obligation to proceed with services during these early discussions.

Step 2: Data Generation & Processing – Metabolon uses advanced technology, robust standard operating procedures (SOPs), and highly reproducible methods to identify and quantify all metabolites from nearly any sample type. In all service offerings, data are expertly processed and rigorously quality controlled to ensure optimal results.  Learn more about specific data-generation and processing methods in Services section below.

Step 3: Expert Interpretation – Once metabolites are identified and quantified, Metabolon provides the client with expert biological interpretation by a team of experienced scientists, statisticians and bioinformaticians. Deliverables vary based on the services selected, but typically include a comprehensive report (complete with heat map displays, raw data, relevant statistics, literature support, and suggestions for next steps) and a PowerPoint file summarizing findings. All offerings also include access to the MetaboLync™ Client Portal, which allows users the option to share their study data with others at their home institution and features a suite of data visualization and navigation tools.

Metabolon Services:

  • Broad metabolite coverage, discovery and interpretation of known metabolites and their pathways (mView™)
    • Samples screened using ultrahigh performance liquid chromatography (including reversed-phase and hydrophilic interaction chromatography (HILIC) methods) with tandem mass spectrometry (UHPLC/MS/MS) and gas chromatography/mass spectrometry (GC/MS)
    • Metabolites are rapidly, automatically identified using an extensive biochemical reference library of more than 4,000 known metabolites (additional screening against our library of 9,000+ novel metabolites available at an extra charge).
    • Advanced bioinformatics applied to refine data and eliminate noise, including component construction, chromatographic peak alignment tools, and spectral library matching of analytical signatures for compound identification.
    • Data subjected to rigorous control procedures including manual data curation, power calculations and statistical analysis of P-value and Q-Value, to eliminate false positives and ensure accurate identification and quantitation of metabolites. This offering is a flagship solution for academic researchers.

 

  • Focused panels profiling metabolites within select metabolic classes (TrueMass® Focused Panels)
    • Samples screened using GC-MS, LC-MS or GC-FID methods dependent on specific analytes to be measured. The absolute concentration of each analyte is determined by comparing the peak area to that of the appropriate internal standard.
    • Capable of screening all 450 complex lipid metabolites (TrueMass® Lipomics), or focusing on 10 to 40 metabolites within a given class for a more in-depth understanding of a particular pathway or system of biology, such as sterol, bile acid, acylcarnitine or ceramide biology.
    • Study results are provided via Surveyor® client tools, a secure online application that provides pathway visualization and data interpretation support.
    • Useful for hypothesis refinement and validation, as well as clinical monitoring.
    • Custom panel development is also available (see TrueMass Targeted Assays).

 

  • Customized profiling for select metabolites (TrueMass® Targeted Assays):
    • Custom assays or panels of one to ten metabolites developed by a team of highly-qualified Ph.D. Bioanalytical Chemists with decades of experience in bioanalysis and assay development.
    • LC-MS/MS assay development and sample analysis has been standardized on ABSciex Qtrap 5500 mass spectrometry with UPLC input. GC-MS/MS assay development is carried out on a Thermo-Finnigan TSQ XLS GC-MS/MS system.
    • Assays are validated assessing linearity, LLOQ precision, accuracy, QC precision, carry-over, recovery and stability for each analyte.
    • Useful for novel discoveries, hypothesis refinement, validation, and clinical monitoring.

 

  • Quantose IR™ – Laboratory developed test  for measuring insulin resistance (IR)
    • Sensitive, accurate and effective clinically-validated blood test for measuring insulin resistance.
    • Allows for precise identification of IR status of subjects in any study where subjects experience a change in weight or glycemic status.
    • Tracks improvements in insulin sensitivity when HbA1C does not.
    • Useful for determining drug treatment and diet effects in cardiovascular, metabolic disease, and other trial populations

 

  • Comprehensive assessment of the entire Metabolome, including complex lipids (TrueVision™)
    • Samples screened using ultrahigh performance liquid chromatography (including reversed-phase and hydrophilic interaction chromatography (HILIC) methods) with tandem mass spectrometry (UHPLC/MS/MS) and gas chromatography/mass spectrometry (GC/MS). A variety of mass spectrometers are employed, including accurate mass systems equipped with both ESI and APCI probes.
    • Metabolites are rapidly, automatically identified using an extensive biochemical reference library containing more than 14,000 known and novel metabolites, including 450 individual lipid metabolites across 10 lipid classes.
    • Advanced bioinformatics applied to refine data and eliminate noise, including component construction, chromatographic peak alignment tools, and spectral library matching of analytical signatures for compound identification.
    • Data subjected to rigorous control procedures including manual data curation, power calculations and statistical anlaysis of P-value and Q-Value, to eliminate false positives and ensure accurate identification and quantitiation of metabolites.
